antihypertensive effects by inhibiting Angiotensin <br />
<br />
<br />
<br />
Converting Enzyme (ACE) and diuretics effect. <br />
<br />
<br />
<br />
The mechanism of action of binahong and <br />
<br />
<br />
<br />
tempuyung plants againts angiotensin II receptors <br />
<br />
<br />
<br />
is unknown. This study aims to determine the <br />
<br />
<br />
<br />
antihypertensive effect of ethanolic extract of <br />
<br />
<br />
<br />
binahong leaf (EEBL), ethanolic extract of <br />
<br />
<br />
<br />
tempuyung leaf (EETL), and their combination in <br />
<br />
<br />
<br />
angiotensin II-induced hypertensive animal <br />
<br />
<br />
<br />
model. The blood pressure was measured noninvasively using CODA® tail-cuff blood pressure <br />
<br />
<br />
<br />
system. The study used 27 male Wistar rats aged <br />
<br />
<br />
<br />
4-5 months, 250-350 grams devided into nine <br />
<br />
<br />
<br />
groups; normal group, sick group, valsartan <br />
<br />
<br />
<br />
group, EEBL 50 mg/ kg bw group, EEBL 100 <br />
<br />
<br />
<br />
mg/ kg bw group, EETL 50 mg/ kg bw group, <br />
<br />
<br />
<br />
EETL 100 mg/ kg bw group, combination of <br />
<br />
<br />
<br />
EEBL –EETL (25 mg/ kg bw : 25 mg/ kg bw) <br />

until normal blood pressure obtained. Initial <br />

blood pressure was measured as T0, then rats <br />

were given orally: 0.5% Na-CMC solution for the <br />
<br />
<br />
<br />
normal and sick groups, EEBL 50 mg / kg bw, <br />
<br />
<br />
<br />
EEBL 100 mg / kg bw, EETL 50 mg / kg bw, <br />
<br />
<br />
<br />
EETL 100 mg / kg bw, combination EEBL-EETL <br />
<br />
<br />
<br />
(25 mg / kg bb: 25mg / kg bb), and a combination <br />
<br />
<br />
<br />
of EEBL-EETL (50 mg / kg bw: 50 mg / kg bw). <br />
<br />

One hour after given extract, angiotensin II 100 <br />
<br />
<br />
µg / kg bw diluted in 0.9% NaCl solution was <br />
<br />
<br />
<br />
injected intraperitoneally to all test groups except <br />
<br />
<br />
<br />
for normal group was injected with 0.9% NaCl <br />
<br />
<br />
<br />
solution. Two minutes after the injection, blood <br />
<br />
<br />
<br />
pressure was measured again (T1). The result <br />
<br />
<br />
<br />
shown that EEBL 50 mg/ kg bw group, EEBL <br />
<br />
<br />
<br />
100 mg/ kg bw group, EETL 100 mg/ kg bw <br />
<br />
<br />
<br />
group, combination of EEBL –EETL (25 mg/ kg <br />
<br />
<br />
<br />
bw : 25 mg/ kg bw) group, combination of EEBL- <br />

<br />
<br />
EETL (50 mg/ kg bw : 50 mg/ kg bw) group <br />

provide prevention an increase of systolic blood <br />
<br />
<br />
<br />
pressure (SBP) and diastolic blood pressure <br />
<br />
<br />
<br />
(DBP). EEBT 100 mg / kg bw and a combination <br />
<br />
<br />
<br />
of EEBL-EETL (25 mg / kg bw : 25 mg / kg bw) <br />
<br />
<br />
<br />
can significantly prevent an increase of SBP and <br />
<br />
<br />
<br />
DBP (p <0.05) with inhibition of TDS 48.2% and <br />
<br />
<br />
<br />
54.6%, the inhibition of TDD is 60.7% and <br />
<br />
<br />
<br />
67.6%. Based on the results above, it can be <br />
<br />
<br />
<br />
concluded that EETL 100 mg / kg bw and <br />
<br />
<br />
<br />
combination of EEBL-EETL (25 mg / kg bw : 25 <br />
<br />
<br />
<br />
mg / kg bw can significantly reduce the action of <br />
<br />
<br />
<br />
angiotensin II (p<0.05). <br />
