Summary: |
Instrumentation system has been developed to identify cardiac activity in mice (Mus musculus). Mice are one of the pre-clinical animals which necessary in research and further development of medicine. Purpose of the test is to monitor the pattern and rhythm change of electrical signal which generate from the mice’s heartbeat as called electrocardiogram (ECG). In Indonesia, the existence of ECG instruments specified for mice is very rare and relatively more expensive than ECG for humans.
Characteristics of the ECG signal generates from mice are different from human, which determined by the faster heartbeat and the smaller anatomical size. Those characteristics are the substantial consideration in the development of this prototype. The location and depth of needle electrodes and also anatomical information of the mouse are determined by 3D micro-CT. The output of microCT is an anatomical axial image of mice. According to axial micro-CT image analysis from the sample of mice, the depth of needle electrodes should not be more than 0.9 mm and the distance of needle electrodes which placed on the heart to the armpit must be around 0.6533 mm. The image analysis constructs from the sample mice’s heart region.
The mouse ECG signal amplifier system uses the openBCI ganglion board which consists of 4 measurement channels with programmable gain. To reduce the source of interference during ECG measurements in mice, the Wim-ECG system is created to reduce motion artifacts during measurement, facilitate consistent electrode injection in mice and provide instrumentation system maintenance. The software in this study is developed for the purpose of wireless interface communication, on-line data monitoring and acquisition and further signal processing.
Evaluation of ECG signal measurement is carried out in 3 different mice, which are first a male mouse weighted 50 grams, second a male mouse weighted 31 grams and third a female mouse weighted 30.3 grams. The three mice are given ketamine drugs with different dose of 0.1 ml/gr, 0.06 ml/gr and 0.0606 ml/gr respectively. The results show an average RR interval at 0.094as, an average QRS width at 0.043s and an average heart rate at 529.85 bpm from the first mouse. The second mouse shows an average of RR interval at 0.131s, an average of QRS width at 0,041s and an average heartbeat rate at 510.33 bpm. The third mouse shows an average of RR interval at 0.126s, an average of QRS width at 0,046s and an average heartbeat rate at 495.81 bpm.
|
