Most of drugs were obtained from plants. Indonesia is recognized as the country having richest biodiversity and the great number of medicinal plants in the world. Indonesia has 7000 species of medicinal plants and 940 species of them have been identified to have medicinal properties. One of plants known to have having various pharmacological properties and it is still reported limitedly is Garuga genus. Garuga is one of genus belongs to Burseraceae family and it has 615 species spreading across Indonesia, Malaysia, Philippines, Papua New Guinea, India, Bangladesh, North Australia and America. This genus is known to produce cyclic diarylheptanoids compounds as major compounds that showed various bioactivities, such as anticancer, cytotoxic, antibacterial, antioxidant, antidiabetic and antiinflammatory. One of Garuga species growing in Indonesia is Garuga floribunda. From previous research it was reported that only one compound isolated from stem bark of G. floribunda. Its leaves have been reported to contain flavonoids, polyphenols, anthraquinones and terpenoids, qualitatively. Therefore, in this research isolation of secondary metabolites from G. floribunda stem bark and structures characterization of isolated compounds have been conducted. The isolation of secondary metabolites have been carried out by several steps. First, the stem bark powder was extracted in room temperature (maceration) by using acetone to get acetone extract. Next, the acetone extract was further fractionated and purified by using several chromatography methods, such as Vacuum Liquid Chromatography (VLC), Radial Chromatography (RC) and Gravitational Column Chromatography (GCC). Determination of structure of isolated compounds has been conducted according to spectroscopic data, i.e. 1D-NMR (1H-NMR and 13C-NMR) and 2D-NMR (HSQC and HMBC). Four pure compounds have been isolated from acetone extract of G. floribunda stem bark. They have been characterized and suggested as cyclic diarylheptanoids, i.e. one new is biphenyl type cyclic diarylheptanoid namely Gf-3 along with three known diphenyl ether type cyclic diarylheptanoids identified as garugamblin I, 9’-desmethylgarugamblin I, and garuganin I. Garugamblin I has been reported from stem bark of G. floribunda and its varieties (G. gamblei), 9’-desmethylgarugamblin I has been isolated from G. pinnata stem bark, while garuganin I has been found in stembark of G. pinnata and G. gamblei. text |
