Its population has been decreasing due to overexploitation and habitat fragmentation. Therefore, effective micropropagation protocol has to be developed in order to conserve this orchid species. The purpose of this study was to determine optimum combination of 6-benzylaminopurine (BAP) and/or coconut water (CW) for axillary shoot induction and compare two media containing banana extract or coconut water for Dendrobium leporinum callus and PLB differentiation into plantlets. In this study, small plantlets were used as explants and cultured in liquid Vacin-Went (VW) medium supplemented with 20 g/L sucrose, 5 mg/L BAP, and 15% (v/v) CW in rotary shaker for nine weeks. Axillary shoot-induced explants were transferred to solid VW medium supplemented with BAP (0, 5, or 10 mg/L) and with or without addition of 15% (v/v) CW. Calli and PLBs formed on explants were transferred to solid half-strength Murashige-Skoog medium supplemented with 20 g/L sucrose and 50 g/L ambon banana or 30 g/L sucrose, 10% (v/v) CW, and 2 mg/L BAP. Axillary shoot multiplication and callus-PLB differentiation into plantlets were observed. Data analysis were done with Kruskal-Wallis test and Dunn's post hoc test (p<0,05) for both axillary shoot multiplication and callus-PLB differentiation. Results showed that highest axillary shoot multiplication was observed in solid VW medium without CW and BAP with average of 2,86 ± 1,08 shoots/explant while solid half-strength MS medium supplemented with 20 g/L sucrose and 50 g/L ambon banana is effective for callus and PLB differentiation into plantlets with average percentage of 56 ± 0,37%. Thus, optimal medium for axillary shoot multiplication is VW medium without CW and BAP, whereas half-strength MS medium supplemented with 20 g/L sucrose and 50 g/L ambon banana is effective for callus-PLB differentiation into plantlets. It is concluded that VW medium without CW and BAP is effective for axillary shoot multiplication and solid half-strength MS medium supplemented with 20 g/L sucrose and 50 g/L ambon banana is effective for callus and PLB differentiation into plantlets.<p align="justify"> text |
