Summary: | <p align="justify"> Palm oil is one of the big foreign exchange source in Indonesia. Furthermore, Indonesia becomes the top of CPO (Crude Palm Oil) producer in the world. Oil’s quality is one of important factors in CPO production and it can be evaluated based on acidity level in palm oil by measuring the FFA (Free Fatty Acid) content. The higher FFA value in palm oil, the lower its quality becomes. Moreover, palm oil with FFA content >5% is considered to be unfit for human consumption. Validating SSR marker, mEgCIR_LIP03, as screening tool for palm oil with low FFA content becomes objective of this research. This research has 4 main stages, those are protocol optimization, phenotyping, genotyping and marker validation. Protocol optimization of oil extraction refers to two titration method literatures. The most consistent oil extraction protocol is using isopropanol. In the phenotyping stage, FFA level analysis is obtained from 5 population and 27 progeny. One-way ANOVA statistic analysis shows p-value=0.01 from population and p-value=0.384 from progeny. P-value results indicate significant differences level of FFA enhancement from population, but from progeny there are no vivid difference level. The next step is genotyping. In this step, DNA samples are amplified by mEgCIR_LIP03 primer and the amplicon are visualized using Fragment Analyzer to measure formed allele size. Genotyping results denote that allele with size 262bp, 264bp and 266bp are allele size that appear frequently in all population. The final step is mEgCIR_LIP03 marker validation. In this step, each individual is grouped based on its formed allele size with its FFA content then its analyzed using one-way ANOVA method. There are no significant differences between alleles which are tested based on its FFA content. Distribution of FFA level enhancement shows that all individual are skewed on the left. It shows slight FFA content enhancement, and also indicate that mEgCIR_LIP03 marker can not distinguish FFA content which are skewed on the left, that have very low FFA enhancement. In the other words, mEgCIR_LIP03 marker is not powerful enough as a screening tool for palm oil fruits that have very low FFA enhancement. Even , from this research, it can be discovered plant materials which have low FFA enhancement that can be used for breeding with low FFA variety. Further research is needed to discover more appropriate genetic maker. <p align="justify"> |
