(MO) nanoparticle coated with sodium alginate, Eudragit S100, and <br /> <br /> Eudragit FS 30D. Nanoparticle was prepared by ionotropic gelation method using chitosan polymer <br /> <br /> as matrix and sodium tripolyphosphate as cross-linker. The particle size and polydispersity index <br /> <br /> were characterized using the DelsaTM NanoC (Beckman Coulter) instrument, the entrapment <br /> <br /> efficiency was determined using the Bradford method (for BSA) and the Chang method (for extract), <br /> <br /> the release profile was evaluated in vitro, and the immune response was evaluated in vivo in BALB/c <br /> <br /> strain mice by determining antibody titer using the immunodifusion method. The results of in vitro <br /> <br /> showed Eudragit S100 is the best polymer in preventing BSA release in gastric pH (1,2) and the <br /> <br /> fastest polymer in releasing BSA at pH ileum (7,4). The result of in vivo showed BSA-MO (100 μg/mL) <br /> <br /> coated with sodium alginate induce the highest humoral immune response base on antibody titer <br /> <br /> (1:128) compared to other orally administered nanoparticle although still lower than parenteral <br /> <br /> administration (1:256). From these results obtained that oral vaccine preparation containing BSA <br /> <br /> and Moringa leaf water extract nanoparticle coated with sodium alginate capable of inducing a <br /> <br /> highest humoral immune response compared with other nanoparticle preparations although <br /> <br /> immunogenicity is lower than parenteral administration. <br /> text |
