Summary:Indonesia is a country where infection is still a common disease. One of the bacteria that can <br /> <br /> cause infection and a common one is Escherichia coli. This bacteria can be treated by beta-lactam <br /> <br /> antibiotics. Third generation of sephalosporin, ceftriaxone, is antibiotic that included in the <br /> <br /> guideline for E. coli infection treatment. Excessive use and misuse of antibiotics can lead to <br /> <br /> bacteria resistance of certain antibiotics. The mechanism can be making beta-lactamase enzymes. <br /> <br /> The purpose of this research is to determine the subgenotipe of blaCTX-M gene from the clinical <br /> <br /> isolate of E. coli that resistance to ceftriaxone and get treatment recommendation for patient that <br /> <br /> infected by E. coli that resistance to ceftriaxone based to the result of this research. The blaCTX-M <br /> <br /> gene in clinical isolates of E. coli that resistance to ceftriaxone was checked by Polymerase Chain <br /> <br /> Reaction (PCR) with the colony as DNA template. The PCR result then visualized by agarose <br /> <br /> electrophoresis to see the fluoroscence on the electrophoregram that show the blaCTX-M gene. <br /> <br /> Samples that contain blaCTX-M gene was sequenced. The sequences were aligned to blaCTX-M gene <br /> <br /> in software GenBank and homologous/identity persentages were determined to get the <br /> <br /> subgenotipe of blaCTX-M gene. From 15 samples that have blaCTX-M gene, 5 of the sample has blaCTX- <br /> <br /> M-15 and was chosen to be sequenced. All of them contained blaCTX-M-15 gene with 100% <br /> <br /> homologous. E. coli that contain blaCTX-M-15 resistant to various beta-lactam antibiotics. Antibiotics <br /> <br /> recommendation for treat E. coli that contain blaCTX-M-15 is carbapenem subgroup and betalactams with tazobactam combination. <br />
