Based on the previous research, Gnetin C is identified as the major active compound in <br /> <br /> melinjo seed extract from both the endosperm and shell. Gnetin C is a stylbenoid dimer and has <br /> <br /> demonstrated inhibitory activity on tyrosinase enzyme. This research is aimed to study the influence of <br /> <br /> particle size of the simplisia of young melinjo seed endosperm on the tyrosinase inhibitory bioactivity. <br /> <br /> Particle size reduction of young melinjo endosperm was performed by ball milling technique with <br /> <br /> various milling time of 0.5; 1; 1.5; and 2 hours. Physical characterization of melinjo fine powder was <br /> <br /> carried out including particle size by Particle Size Analyzer and the particle morphology using Scanning <br /> <br /> Electron Microscope. The PSA data showed that the optimum milling time to produce particle with size <br /> <br /> of 650 nm was 2 hours. However, the highest total phenol and tyrosinase inhibitory activity was <br /> <br /> demonstrated when the milling time was 1.5 hours, accounted as 11.98 ± 0.02 GAE µg/mg dried sample <br /> <br /> and 35.46 ± 1.29 % respectively. HPLC analysis on extract powder milled for 1.5 hours showed that the <br /> <br /> content of Gnetin C was about 0.54%. Focusing on the tyrosinase inhibitory activity data, combination <br /> <br /> of phenol compounds, Resveratrol, Gnetin C and Gnemonoside A is suggested to contribute on the <br /> <br /> activity. <br /> text |
