Summary:<b>Abstract: </b><br> A study on the effect of elicitor derived from <u>Verticillium dahliae</u> and <u>Rhizoctonia solani</u> on gosipol production of <u>Gossypium hirsutum</u> cell aggregates have been conducted. Cell aggregates culture were initiated from friable callus induced from <u>G. hirsutum</u> cotyledone on Linsmaier & Skoog (LS) medium. Cell aggregates culture of <u>G. hirsutum</u> grew optimally in LS medium containing 10<sup>-7</sup> M naphthalene acetic acid (NAA) and 10<sup>-6</sup> M 2,4-dichlorophenoxyacetic acid (2,4-D). Cell aggregates were elicitated with elicitor derived from autoclaved <u>V. dahliae</u> and <u>R. solani</u>. The concentration used were 20, 40, 60, and 80 µg DW/mL. The harvesting times were 2, 4, and 6 days. Gossypol was analysed qualitatively and quantitatively using High Performance Liquid Chromatography (HPLC). Elicitation by <u>V. dahliae</u> and <u>R. solani</u> increased gossypol production in <u>G. hirsutum</u> cell aggregates. The optimum concentration of <u>V. dahliae</u> elicitor which elicited highest gossypol production was 60 µg DW/mL, four days after elicitation (41.7 ± 1.2 µg/mL, control 22.6 ± 1.2 µg/mL), whilst that from <u>R. solani</u> was 20 µg DW/mL, in second days after elicitation (104.0 ± 10.7 µg/mL control 21.1 ± 2.2 µg/mL)
