The objectives of this research were to study <br /> <br /> antioxidant activity of four parts of pemphis i.e root bark, flower, stem bark, and leaves by <br /> <br /> DPPH and FRAP methods, to determine total phenol and flavonoid, to analyze the <br /> <br /> correlation between total phenolic and flavonoid content towards IC50 DPPH and EC50 <br /> <br /> FRAP, also correlation between IC50 DPPH and EC50 FRAP. Methods: Extraction was <br /> <br /> conducted by reflux using gradient polarity solvents. The extracts were evaporated using <br /> <br /> rotary vacuum evaporator. Determination of IC50 of DPPH scavenging activity and EC50 of <br /> <br /> FRAP capacity, phenolic and flavonoid content were performed by ultraviolet-visible <br /> <br /> spectrophotometry. Correlation between total phenolic and flavonoid content with IC50 <br /> <br /> DPPH and EC50 FRAP, also correlation between two antioxidant testing methods were <br /> <br /> analyzed by Pearson’s method. Results: All ethyl acetate and ethanol extracts of pemphis <br /> <br /> root bark, flower, stem bark and leaves had IC50 DPPH < 50 μg/mL. The ethyl acetate root <br /> <br /> bark extract of pemphis by DPPH method (IC50 0.41 μg/mL) and the ethanol root bark <br /> <br /> extract by FRAP method (EC50 3.11 μg/mL) had the highest antioxidant activity. The ethyl <br /> <br /> acetate pemphis flower extract had the highest total phenolic content (19.75 g GAE/100 g) <br /> <br /> and ethyl acetate pemphis leaves extract had the highest total flavonoid content (17.49 g <br /> <br /> QE/100 g). Total phenolic content in pemphis leaves extract had negative and significant <br /> <br /> correlation with its EC50 of FRAP. IC50 of DPPH and EC50 of FRAP were linear in all <br /> <br /> extracts. Conclusions: All ethyl acetate and ethanol extracts of root bark, flower, stem bark, <br /> <br /> and leaves of pemphis were very strong antioxidant. Phenolic compounds were the main <br /> <br /> contributor of antioxidant activity of pemphis leaves extract by FRAP method. DPPH and <br /> <br /> FRAP methods gave linear result in antioxidant activity of all pemphis extracts. <br /> text |
