Summary:The HPLC (High Performance Liquid Chromatography) separation method using silica columns <br /> <br /> modified by amine are widely used, especially in the separation of sugar mixture. In this research, <br /> <br /> silica fume column with particle size 325 - 400 mesh have been made. Silica fume was activated with <br /> <br /> standard piranha solution, then reacted with 3-APTES, whose diluted in methanol and demineralized <br /> <br /> water. FTIR characterization using DRS method showed the presence of a new peak at wavenumber <br /> <br /> 1650 - 1500 cm-1. That peak is refer to amine functional group and can indicated that the <br /> <br /> modification was success. Silica fume modified by amine then tested for separation performance <br /> <br /> towards fructose and maltose mixture. Measurements were carried out by the FIA-UV method (Flow <br /> <br /> Injection Analysis-Ultraviolet) at wavelength 282 nm. Spectrum analysis showed the separation <br /> <br /> between fructose and maltose is happened. Silica fume after modified by amine also showed slimer <br /> <br /> peaks than silica fume before modified.
