palm oil and jojoba oil. The composition of jojoba oil is similar to human sebum’s composition which <br />
<br />
produced by sebaceous gland. The aim of this research is to develop a method in determining the <br />
<br />
concentration of β-carotene present in jojoba oil (Simmondsia chinensis) by transesterification <br />
<br />
reaction using visible spectrophotometer method and determined the concentration of β-carotene in <br />
<br />
jojoba oil. The method was developed to obtain the highest levels of β-carotene based on the <br />
<br />
optimum condition in the preliminary reaction. Transesterification reaction used 1 grams of NaOH and <br />
<br />
54 grams of metanol to produced 77,302 grams of the highest level fatty acid methyl ester (FAME). <br />
<br />
Validation method was performed to determine of linearity, limit of detection, limit of quantification <br />
<br />
,accuracy, and precision. Based on the results of validation method, linearity test obtained r of 0.9995 <br />
<br />
and r <br />

of 0.9992; accuracy test %recovery of 90.094 – 101.022 %; limit of detection is 0.89 μg/mL and <br />
<br />
limit of quantification is 2.964 μg/mL, precision test obtained relative standard deviation of 4.687; <br />
<br />
1.246; 0.859 % for intraday test and 4.687 and 1.837% for interday test. Method is able to determine <br />
β-carotene in jojoba oil and the level β-carotene in jojoba oil is 155,641 μg/mL. <br />
