The aim of this research is determine the potential of Neurospora sitophila on tobacco leaves for increasing the recovery of nicotine compounds. Neurospora sitophila has an ability to synthesize and secrete cellulase enzymes that include in cellulosic hydrolysis and various hydrolytic enzymes to degrade lignin and hemicellulose. In this study, Neurospora sitophila was used as a biological agent in the pretreatment process of lignin degradation. The method for the pretreatment process is solid phase fermentation (SSF). This method uses the ratio of tobacco leaf powder and inoculum of 3:4 (b / v). Inoculum is prepared in a medium potato dextrose broth (PDB) by adding 20% (v / v) medium of Toyama's Medium Solution (TMS) as a source of phosphate fungus Neurospora sitophila. The optimum time to inoculate the Neurospora sitophila on the substrate is on the third day because on that day Neurospora sitophila has a cell concentration of 1.47x107 cells / ml with %viability of 37.08%. This optimum condition is expected to improve SSF process in degrading the lignin of tobacco leaf powder. Fermentation is done for 4 days with variation of fermentation time, that is 1, 2, 3, and 4. The fermentation process in tobacco leaf can increase the oil of nicotine extract by 3.34% (b / b) and increase the nicotine compound by 25,47% from 67.38% to 90.4% with degradation of hemicellulose become 32.93%, cellulose become 12.58%, and lignin become 51.84%. text
