Water <br /> <br /> Spinach and Gotu Kola’s leaves have been used traditionally to treat insomnia. The purpose of this <br /> <br /> research is to determine the most potent extract with sedative hypnotics effect from extract of <br /> <br /> Gotu Kola and Water Spinach, and to determine the combination of extracts that could give the <br /> <br /> most potent. The extracts were obtained by maceration with ethanol and decoction water. The <br /> <br /> study used mice divided into 10 groups. Diazepam was used as reference, and 0,5% CMC-Na was <br /> <br /> used as vehicle. Test group mice received one of these treatment: water extract of spinach at 100 <br /> <br /> and 200 mg / kg BW, ethanolic extract of Spinach at 100 and 200 mg / kg BW, water extract of <br /> <br /> Gotu Kola at 100 and 200 mg / kg BW, and ethanolic extract of Gotu Kola at 100 and 200 mg / kg <br /> <br /> BW. The hypnotic effect test was tested by measuring the parameters of sleep latency, sleep <br /> <br /> duration, sleep efficiency, and sleep cycle in mice previously induced with ketamine (100 mg / kg <br /> <br /> BW). In addition, other parameters such as the behaviour and the sedation rating scale, to see if <br /> <br /> there were any sedative effects, were tested. Data were analyzed statistically using One way <br /> <br /> ANOVA and Post Hoc Test (LSD). The results showed that there was no significant difference <br /> <br /> between water extract and ethanolic extract of Water Spinach and Gotu Kola in all hypnotic <br /> <br /> parameters tested. However, with regard to sleep efficiency, water extract of Spinach and Gotu <br /> <br /> Kola at 100 mg/kg BW had higher efficiency than the ethanolic extract. Therefore, these extracts <br /> <br /> were choosen for further combination study. It was shown that combinations in the ratio of 1:3 <br /> <br /> and 3:1 produced higher sleep efficiency, faster sleep cycle and faster reduction of motoric <br /> <br /> activity than the extract alone. Furthermore, combination of Gotu Kola at 25 mg/kg BW and <br /> <br /> Water Spinach at 75 mg/kg BW had synergistic activity compared to the individual extract. <br /> text
