Summary: | The research on photosynthetic pigments of the tropical marine microalgaes as antioxidants, solar cells, and sunscreen has been perfomed in the Biochemistry Laboratory, Institut Teknologi Bandung. However, research on the modification of fucoxanthin Cyclotella striata strain TBI into novel chemical compounds has not been reported yet. The purpose of this study was to modify fucoxanthin C. striata strain TBI through the metathesis olefin method. The methods included large-scale biomass preparation, pigment extraction using acetone and fucoxanthin purification using silica gel column, fucoxanthin modification through the metathesis olefin method, and fucoxanthin as well as the products characterization using TLC, UV-VIS spectrophotometer, HPLC, and FTIR. The productivity of C. striata strain TBI obtained from an initial density of 2.65×105 cells/mL was 208 mg wet biomass (L culture)–1 d–1, or equivalent to 13.4 mg dried biomass (L culture)–1 day–1. The Rf value fucoxanthin from C. striata TBI strain was 0.23, and the fucoxanthin shows fingerprint absorption at 447 and 470 nm. The retention time of fucoxanthin was 5.332 min (Shim-pack Vp-ODS, methanol:acetonitrile (80:20)). The vibration of atomic bonds in the fucoxanthin molecule was detected at wave numbers of 3427 (–OH), 2924 (–CH alkane), 1928 (C=C=C), 1730 (–C=O ketone), 1653 (–C=C), 1455-1366 (–CH alkane), and 1029 (–C–O ester) cm–1. Fucoxanthin C. striata strain TBI obtained from 1 g dried biomass was 6 mg, or equivalent to the productivity of 0.64 μg fucoxanthin (L culture)–1 d–1. The five products of fucoxanthin C. striata strain TBI self-methatesis identified with TLC had a Rf value of 0.07, 0.16, 0.30, 0.36, and 0.41, respectively. |
