oxidized, thus a process is needed to increase particle stability, namely coating the particle with chitosan. <br />
<br />
In this research, Fe3O4 modified with chitosan (Fe3O4-C) was synthesized and its affinity with protein was <br />
<br />
observed. The protein used was peroxidase enzyme from radish. Synthesis of Fe3O4 dan Fe3O4-C was done <br />
<br />
using co-precipitation method. Chitosan was crosslinked on the surface of Fe3O4 with a combination of <br />
<br />
tripolyphosphate/sulfate. Characterization of both particles included particle weighing, magnetic <br />
<br />
properties, infrared spectrum, and their interaction with proteins. The results showed that the weights of <br />
<br />
Fe3O4 and Fe3O4-C were 1.20 ± 0.02 gram dan 0.11 ± 0.003 gram, respectively. Precipitation times of Fe3O4 <br />
<br />
and Fe3O4-C with and without magnets were different but both had magnetic properties. The results of <br />
<br />
interaction between particles and protein showed a decrease of the total protein amount and peroxidase <br />
<br />
activity. The largest decrease of total protein amount occurred at pH 7 for 30 minutes, which was 55% <br />
<br />
with peroxidase activity 0.01 U/mg. The particle resulted from the interaction between particles and <br />
<br />
protein solution (Fe3O4-P dan Fe3O4-CP) had peroxidase activity and magnetic properties, hence showing <br />
<br />
that Fe3O4 and Fe3O4-C can be used for enzyme immobilization. <br />
