The aim of this study is to evaluate the best condition to induce transgenic hairy roots as alternative method means for saponin production in T. paniculatum. Two different strain A. rhizogenes LBA9402/12 and ATCC 15834 were used for induction of hairy roots in in vivo and in vitro leaf explant. The effect of bacterial density, incubation period, and the addition of acetocyringone were evaluated to obtain the optimum condition for transformation. The consentration of saponin was measured using spectrophotometric method based on the standard calibration curve of oleanolic acid. The highest frequency of transformation (90%) was generated on in vivo leaf explant following 5 minutes incubation at an OD = 1 and in the absence of acetocyringone. Saponin analysis showed that hairy root cultures accumulate higher saponin content than non-transgenic root. The content of saponin from hairy root, in vivo and in vitro root extract was as follow 2,01% (w/w) ; 1,56% ; and 1,43%. Thus, hairy root offer a good system for modulation of saponin production in the medicinal plant T. paniculatum. text |
