Furosemide is loop

diuretic which is recommended for hypertension accompanied by edema. Furosemide is widely

available in oral dosage form but has many variation and shows a low bioavailability, 60%. Due to

that reason, a furosemide patch was developed in this current study. First, furosemid gel was

prepared using polymer, furosemide, furosemid solvent, plasticizers and penetration enhancers. The

gel was dried at 60 C for 4 hours, then polyisobutylene solution (2.5% in n-hexane) was added on

the top of the film as an adhesive layer. Evaluation of the patch showed that average weight and

average content of furosemide patch were 1.643± 0.009 g and 98.70 ±0.46%, respectively. Tensile

strength and elongation percent of furosemide patch were 0.097 ± 0.003 kg/mm

2

and 4.555±

0.055%, respectively. In vitro skin diffusion test was performed using flow through Franz diffusion

cell and Phyton reticulatus skin used as a membrane. Penetration percentage of furosemide from

patch was 60.02± 2.65% in 24 hours. Irritation test was performed in rabbit's back skin. Stability test

was done in 30 days at room temperature and 40 °C and the results showed a good physical stability,

although there was a tendency of content decrease. Furosemide patch showed uniform weight and

content, its released followed Higuchi kinetic and did not showed irritation on the skin.
text
