Summary:Cyclotella striata TBI is a type of tropical marine diatom producing highly lipids. One of the <br /> <br /> lipid is polyunsaturated fatty acids (PUFAs). The aim of this research was to isolate PUFA <br /> <br /> from C. striata TBI and convert PUFA methyl ester into1,4-sikloheksadiena. The research <br /> <br /> stages included indoor seeding C. striata TBI cells, outdoor cultivation of C. striata TBI <br /> <br /> cells, harvesting biomass, isolation and lipid transesterification of C. striata TBI, seperation <br /> <br /> and parsially purification of fatty acid methyl ester, determination of the fatty acid content of <br /> <br /> C. striata TBI, and conversion of PUFA methyl ester using Grubbs II catalyst. The results <br /> <br /> showed that the biomass productivity of C. striata TBI cultured outdoor from three <br /> <br /> cultivations separately was have 13.93, 184, and 155 mg/L culture/d, respectively. <br /> <br /> Meanwhile, the lipid productivity of C. striata TBI was 15 mg/g dried biomass/d. The <br /> <br /> amount of PUFA in total fatty acid C. striata TBI was 20.24% (w/w). Fraction 3 (obtained <br /> <br /> from the seperation of PUFA methyl ester with hexane in 10% acetone) contained PUFA <br /> <br /> methyl esters such as 0.44% (w/w) linolenic acid methyl ester, 6.12% (w/w) eicosapentaenoic <br /> <br /> acid methyl ester, and 0.34% (w/w) docosahexanoic acid methyl ester 0.34%). The <br /> <br /> conversion of PUFA methyl ester using Grubbs II catalyst produced some products. One of <br /> <br /> them identified by GC-MS with a retention time of 2.843 min was 1,4-cyclohexadiene.
