In this study, morphological and genetic analysis of one isolate of SMH microalgae was conducted for the identification and conservation of tropical marine microalgae. The research stages include purification of isolate SMH by gradual dilution, cell propagation, cell morphology analysis with light microscope and electron scanning microscope (SEM), isolation and determination of marker gene sequence (18S rRNA-V4 and LSU D2 / D3). Isolate SMH obtained from Kepulauan Seribu, Untung Java (S 05o 58,193 E 106o 47, 083). The results showed that the pure SMH isolates were able to grow in Walne medium with maximum growth phase on day 12. Cell SMH are round asymmetrical on the surface, green, and 1.83-3.65 ?m in diameter. PCR chromosomes isolates SMH using a specific primer pair of 18S rRNA region V4 and LSU D2/D3 respectively yielded DNA fragments of 0.5 and 0.7 kb size. Nucleotide sequencing results show that 0.5 kb and 0.7 kb amplicon fragments are exactly have 415 bp and 612 bp. Analysis of nucleotide sequences using BLAST (Basic Local Alignment Search Tool) and accompanied by cell morphology analysis showed that SMH isolates had the highest similarity with microalgae genus Chlorella. text
