The study was conducted through cultivation of microalgae P. cruentum in Walne medium and modified medium (NPK 5% w/v), extraction of biopigment by using organic solvents and Phosphate Buffered Saline (PBS) pH 7.4, purification of biopigments by using silica 60G column chromatography, and characterization by using thin layer chromatography (TLC), visible spectrophotometry and sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The results showed that the average cell density of P. cruentum for both Walne and NPK 5% (w/v) medium were 11 x 106 cells/mL culture for 8 d and 10 d cultivation, respectively. Four main visible biopigments from crude extract of acetone were spotted on TLC. Purified biopigments were identified as chlorophyll a (410; 430.8; 662.8 nm), ß-carotene (425; 452; 478, 5 nm) and zeasantin (451; 478 nm). While phycoerythrin was isolated by using PBS pH 7,4 which have characteristic of absorption spectra at 540-570 nm, and size of 18 kDa. The results suggested that the marine microalgae P. cruentum culture can grow in Walne and NPK 5% (w/v) media and produce various types of biopigments. text |
