This plant has a high economic value, especially for silkworms feed and are also used for traditional medicine as a diuretic, expectorant, cure skin diseases, and facilitating the production of breast milk. Morus plants reported to contain secondary metabolites that have potential bioactivity as antitumor drugs, antimicrobial, anti-inflammatory, and antihypertensive. Secondary metabolites in plants are generally so low that it needed another source. Recently, the isolation of secondary metabolites can be performed from the endophytic fungi that live inside plant tissues. Based on literature, a single plant species contained more than one species of endophytic fungi. Some endophytic fungi are involved in the biosynthesis of secondary metabolites of host, so a possible secondary metabolites that are similar to its host. The advantages of this method include that endophytic fungi can be developed in short time, about 2- 3 weeks. In this research, isolation of secondary metabolites from the endophytic fungi at Morus macroura, initiated by inoculation of the ex plant into the solid media Potato Dextrose Agar (PDA). Single colony of the endophytic fungi that have been successfully separated, then subcultured in PDA medium then subcultured into liquid medium Potato Dextrose Broth (PDB) and incubated for 16 days. Furthermore, the endophytic fungi were separated from the media by filtered using a Buchner funnel. Fungi are crushed in a mortar and macerated with methanol, while the media partitioned with ethyl acetate. Secondary metabolite isolation of the media conducted by fractionated the ethyl acetate extract using several chromatographic techniques, such as vacuum liquid chromatography and radial chromatography. The characterizations of isolated compound used were based on spectroscopic data such as NMR, UV-Vis and MS. To determine the bioactivity of isolated compound were obtained, test cells cytotoxic against murine leukemia P-388. Based on spectroscopic data, the pure compounds have been isolated from endophytic fungi M. macroura namely 19,20-epoxy sitokalasin Q which has a highly active cytotoxic bioactivity against murine leukemia cells P-388 with IC50 values of less than 0.1 ?g / mL. text |
