Isolation and Antioxidant Activity Test of Biopigment from Marine Microalgae Thalassiosira sp. Cultivated on Modification Medium

This research has been carried out in isolation of biopigment from marine microalgae Thalassiosira sp. and evaluation of their antioxidant activity by using 2,2-diphenyl-1- phycrylhydrazil (DPPH). The study was conducted through cultivation of microalgae in 3 variations of growth medium (Walne, N...

The study was conducted through cultivation of microalgae in 3 variations of growth medium (Walne, NPK 10 %, modification medium), extraction of biopigments by using acetone p.a., purification of biopigments by using silica column chromatography, and characterization by using Thin Layer Chromatography (TLC)1 Dimensional and 2 Dimensional and visible spectrophotometry. The antioxidant activities against free radical DPPH were tested compare to ascorbic acid as a positive control. The result showed that the modification medium provide optimum biomass density of 3,66 g/L for 6 day cultivation. Five main visible biopigments from crude extract of acetone were spotted on TLC. Purified biopigments were identified as ?-carotene, chlorophyll a, chlorophyllide a, phaeophytin a, and diadinoxantin. The IC50 of ascorbic acid, phaeophytin a, ?-carotene, and diadinoxantin were 5,48 ; 29,01; 8,97 and 5,51 mg/mL respectively. The results suggested that ?-carotene and diadinoxantin from marine microalgae Thalassiosira sp. could be considered as potential source for antioxidant. text
