In medical field, porphyrin derivative is commonly studied as antibacterial and anticancer drugs. Porphyrin structural modification is done to produce a porphyrin derivative that is soluble in water. In this study, structural modification of tetraphenylporphyrin (TPP) was conducted by substituting sulfonate (-SO3H) on all TPP phenyl rings. Characterizations of spectroscopy UV-vis, fluorescence and mass spectrometry, shows that the structure of TPP was successfully modified and meso-tetraphenylsulfonate porphyrin (TPPS4) was produced. Metalloporphyrin of Zn (II) TPPS4 and Cu (II) TPPS4 was synthesized from mixture of TPPS4 solution and metal ion salt (Cu2+ or Zn2+), and pH of the solution was adjusted by using acetate buffer pH 4.8. Formation of both complexes was monitored by measuring the UV-vis spectroscopy and fluorescence. Bioactivity of the TPPS4 solution and metal ions mixture, with stoichiometric ratio 1:5, was studied in plasmid pGEM-T. DNA study shows that solution of the complex compound Zn (II) TPPS4, can cleave the DNA. text
