Summary:<b>ABSTRACT</b><br> Testosterone and oestradiol are male and female primary hormones in the human male and female bodies. In the human male body, the concentration of testosterone is much larger than that of oestradiol and conversely, in the human female body the concentration of oestradiol is much larger than that of testosterone.<p> <br /> Testosterone undergoes metabolism in the liver into androsterone, dehydro-iso-androsterone and ethiocholanolone. The metabolites of testosterone are excreted through urine in the form of water soluble glucuronic acid esters and sulfuric acid esters. Oestradiol undergoes metabolism in the liver into oesterone and oestriol. Oestradiol and its metabolites are also excreted through urine in the form of water soluble glucuronic acid esters and sulfuric acid esters.</p>In the normal individual, testosterone and oestradiol in body play roles in the secondary sexual development in man and woman. The deviation of testosterone and oestradiol from their normal values in man as well as in woman are the first indication of: <p><ol start=\"1\" type=\"a.\"><li>suffering of certain diseases such as; testicular disease, chronic liver disease, androgenic alopecia (baldness in man),in myocard infarction, and others.</li><li>sexual distinction of anybody</li><li>sexual habit,</li><li>certain body condition.</ol></li></p> The aim of this research is to develop a procedure for the determination of testosterone and oestradiol in plasma and urine that is enable to determine both hormones in plasma and urine of man and woman. The acquired concentrations could be used for obtaining the ratio of testosterone to oestradiol (T/E) in man and the ratio of oestradiol to testosterone (E/T) in woman that obviously represent important information for studying physiological and health conditions or hormonal abnormality in man. The testosteron value together with the value of T/E, or the oestradiol value together with the value of E/T, could represent a more significant indication than only that of testosteron or oestradiol.<p>Several methods for the determination of testosterone and oestradiol in body fluids which are currently used are : colorimetric-spectrofotometric methods, spectrofluorometric method, mass-spectrometric method, gas chromatography method, gas chromatography-mass spectrometric method, high performance liquid chromatography (HPLC) method, ion pair high performance liquid chromatography, HPLC-NMR method, radio immuno assay (RIA) method and thin layer chromatography densitometric method, but none of these methods above are used to determine both hormones simultaneously in a biological sample to determine the T/E or E/T values.
