One genus of plants that is potential to be investigated is Morus of Moraceae family which has been reported contains either phenolic compounds, mainly stilbenes, 2-arylbenzofurans, flavonoids, and Diels Alder adducts, or non phenolic compounds, such as terpenoids and steroids. Some of these secondary metabolites have shown important and various bioactivity, such as antitumor, antihypertensive, antimicrobial, anti-inflammatory, and antiviral. Generally, the study of these natural compounds is initiated by the isolation of the natural plants or the plants propagated by tissue culture techniques. The uniqueness of Morus developed by tissue culture techniques is able to produce, especially, phenolic compounds which are more complex and are not commonly found in its natural plants, i.e. the Diels-Alder adducts. Based on these backgrounds, the isolation and characterization of secondary metabolites from Morus shalun root culture were implemented in this research. The aim of this study was to obtain information of the secondary metabolites contained in Morus shalun root cultures for the first time. Root cultures were grown in liquid MS (Murashige- Skoog) media with the addition of hormone IBA (Indole 3-Butiric Acid) 1 ppm. Propagation was done by subculture on root culture aged 4 or 5 weeks for ± 5 months. Isolation was carried out on liquid media (± 7 L) which was extracted with ethyl acetate (EtOAc) (± 5 L). The organic phase (EtOAc) obtained was evaporated with low pressure to get dry EtOAc extract weighing 4.2 g. Isolation and purification using various chromatographic techniques, such as vacuum liquid chromatography, gravity column chromatography, column chromatography sepadhex LH- 20, and radial chromatography yielded three pure compounds. Characterization by spectroscopic data NMR (1D-NMR and 2-D NMR) showed that all three compounds were Diels-Alder adduct derivatives, i.e. kuwanon J, guangsangon E, and calkomorasin which possessed cytotoxicity against murine leukemia P-388 cells with IC50 5,9 ?g/mL, 2,5 ?g/mL, and 1,7 ?g/mL, respectively. text |
