Merr) is one of the most important palawija plants, because of its high protein content and as one of the legumes that generates the secondary metabolite isoflavone. The gene F6H (Flavonoid-6-Hydroxylase) that codes Flavonoid-6-Hydroxylase is known to be involved in the process of glycitein synthesis from liquiritigenin. Increasing the isoflavonoid content of soy seeds is a challenge for farmers in the development of the cultivar, so finding out the mechanism of isoflavonoid synthesis and the relative expression of isoflavonoid genes in the soy seeds are very important to conduct. Glycitein is a soybean isoflavone with a low concentration but is more stable than daidzein and genistein which function as antioxidants for human health. F6H gene codes for F6H enzyme, a cytochrome P450 monooxygenase from the CYP71D subfamily, functiones as a catalyst for the conversion of liquiritigenin into flavanone (6,7,4’-trihydroxyflavanone), which will than be converted into glycitein. The aim of this research was to determine the expression of F6H (F6H1, F6H2, F6H3) gene family in the seed at different growth phases (R5, R6, R7) in several black soybean varieties (UP 106, UP 115, UP 122, UP 157). Expression profile of F6H gene from black soybean seed was carried out using qPCR (Quantitative Real Time PCR) method with F-BOX and MTP housekeeping genes. Of the qPCR data was analyzed using Spearman Rank correlation analysis from statistic software SPSSTM 16.0. The results showed that the expression of F6H family gene from UP 106 and UP 122 was up regulated at R6 phase (F6H1 and F6H3 at Up 106; F6H2 at UP 122). The relative expressions of family gene F6H at R7 phase had been down regulated in all varieties except in UP 115 and 157. The F6H gene correlation analysis between the varieties showed that UP 122 and UP 157 correlated negatively. A gene family expression profile of every phase of growth in every variety showed different patterns. The results showed that all four varieties of black soybean allegedly had genetic varietions that are thought to affect the expression of F6H genes or genes related to isoflavone biosynthesis. |
