Summary:Hepatitis is a disease that caused by Hepatitis B Virus. Patient of Hepatitis can sustain liver cirrhosis and liver cancer that can lead to death. Prevention and treatment of hepatitis B patients are still considered less effective due to the length of time of treatment that can lead to viral resistance and toxicity. Therapeutic and preventative vaccine is proposed as an effective approach to eliminate the virus with antiviral immune response stimulation. HBcAg can bind covalently to MHC class I and may induce the formation of cytotoxic T cells to eliminate hepatitis B-infected hepatocytes cell, and HBsAg can induce humoral immunity effectively. The purpose of this study is to construct a HbsAg-HBcAg fusion gene into pTEV5 expression vector and expressed in Escherichia coli BL21 (DE3). Construction methods generated by ligation HBcAg-HBsAg fusion gene into pTEV5 which had previously been restricted with the enzymes NdeI and EcoRI. Confirmed through PCR method, a method using a restriction enzyme NdeI and EcoRI. HBcAg-HBsAg fusion gene was succesfully constructed in pTEV5. However HBcAg-HBsAg recombinant protein expression in E. coli BL21 (DE3) hasn’t yet expressed. Therefore, in this study it can be concluded that the HBcAg-HBsAg fusion gene has been successfully constructed the expression vector pTEV5.
