Summary:In in vitro regeneration, acclimatization is one of the most critical steps. Enviromental changes from in vitro to ex vitro conditions results low survival and growth of in vitro regenerated plantlets. To reduce tranplantation shock, vesicular arbuscular mycorrhizal (VAM) was applied in order to enhance survival and growth of plantlets. The objective of the research was to evaluate the effects of VAM on growth of in vitro regenerated Physic Nut during the acclimatization phase. In this research a mixed VAM containing Glomus sp, Gigaspora sp and Acaulospora sp in zeolit buffer was used. Inoculation of 5%, 10% and 15% VAM were performed to Physic Nut seedlings from in vitro cultured axillary shoots at the beginning of the acclimatization phase. At this stage, seedlings were transplanted to 90 g pots containing sterilized a soil:sand (1:1) mixture. Physic Nut seedlings from steril sprout and in vivo seed were used as controls. Root length, number of root, fresh weight, shoots length and number of leaf, are used as the growth parameters. Data were collected after six weeks VAM inoculation. Seedlings from in vitro cultured axillary shoots with 15% VAM showed the best respons of survival and growth. Six weeks after VAM inoculation, 66.67% survival percentage was obtained with 33.63 mm root lenght, 5.22 number of root, 0.38 g fresh weight, 38.67 mm shoot lenght and 3.55 number of leaf. Inoculation of 15% VAM in seedlings from in vitro sprout showed 72.92% survival percentage, 32.92 mm root length, 5.00 number of root, 0.40 g fresh weight, 40.90 mm shoot length and 4.10 number of leaf. In vivo seedling with 15% MVA showed 88.89% survival percentage, 7.38 g fresh weight, 100.57 mm shoot leght and 5.43 number of leaf. It was concluded that the VAM biotechnology can be applied for alterate a critical steps in acclimatization phase because the inoculation of VAM significantly improved survival and vegetative growth of in vitro regenerated Physic Nut during the acclimatization phase.
