Production of polysaccharides ?-D-glucan uses two optimization phases. First phase is selecting the best source of corm, such as banana pseudo leaf, cassava and sweet potato. The factor observed on this research is formation of ?-D-glucan polysaccharides. Second phase is modifying the variation of the selected substrat extract variation by adding 300 g / L, 700 g / L and 900 g / L. From the first phase, as observed on day-3, the highest polysaccharides ?-D-glucan rate is found on cassava extract 500 g / L, which amounted to 66.788 %. From the sweet Potato extract the rate obtained on day-3 is 42 %. From extract of banana pseudo leaf the rate obtained is 10.59 % on day-3. Thus, cassava corm extract is the most advantageous media to produce the ?-D-glucan. On the second phase it was observed that on day 3 of the research the highest ?-D-glucan polysaccharides rate is obtained from the concentration of cassava extract 700 g / L and 900 g / L, which amounted to 56.948% and 54.743%. Whilst the lowest ?-D-glucan polysaccharides rate is obtained from cassava extract 900 g / L, which amounted to 41.436%. From the comparison of the highest rate of ?-D-glucan obtained in the second optimization phase, where concentration of cassava extract 700 g / L produces the highest rate of ?-D-glucan amounted to 56.948 %, with the highest rate ?-D-glucan obtained in the first optimization phase, where cassava extract 500 g / L produces the highest rate of ?-D-glucan amounted to 66.78 %, it is deduced that corm of cassava extract 500 g / L is the most advantageous media in ?-D-glucan production from Ganoderma tropicum mushroom. text
