الملخص: | Papain and bromelain are both proteases from papaya and pineapple, respectively. Hydrolysis protein with papain and bromelain produces protein hydrolysate. In this research, papain and bromelain activities were determined using casein as a substrate, and concentration of protein hydrolysate was determined by the method of Lowry. The result showed that specific activity of papain and bromelain was 7.93 and 7.38 U/mg, respectively. The Michaelis-Menten constant (KM) of papain and bromelain obtained from the enzyme kinetic assay was 0.028 and 0.044%, respectively. Molecular weight of papain determined by the SDS-PAGE method was expected to be 39.3 kDa. Meanwhile, bromelain was found to be 2 isoenzymes with Mr 27.6 and 21.8 kDa. The research activities on making protein hydrolysates may be used for creating teaching-model of macromolecular material in high school. Experiences on research works such as isolating crude enzyme from natural products, assaying enzyme kinetics, and determinating macromolecular weight are practical aspects of learning than can be transformed to students, and help them to have better understanding. |
