THE ABILITY OF PSEUDOMONAS AERUGINOSA IN DEGRADATING PARA- NITROFENOL

Bioremediation is a widely used alternative method to overcome the problems of pollution in the environment by exploiting the role of microbes. The compound para-nitrofenol (PNP), also known as 4-nitrofenol or 1-hydroxyl-4-nitrobenzena, abundant side products from fertilizer and medicinal industri...

This compound is dangerous to humans because as it can cause cyanosis and other health disorders. Pseudomonas aeruginosa known as allegedly able to degrade various types of organic and inorganic pollutants, including PNP. This study used Pseudomonas aeruginosa spawn in medium LB (Luria Bertani), which is rich in nutrients, as well as MM (Minimal Medium) by giving pollutant PNP as a pollutant with concentrations of 50, 100, 200, 300, 400 and 500 ppm for LB medium and 20, 40, 60, 80, 100, 120, 140 and 160 for MM. Measurement of bacterial growth pattern was done using UV-VIS spectrophotometer at a wavelength of 600 nm. Optimization of incubation temperature was carried out at 30 oC, 37 oC, 40 oC and 45 oC. Results of PNP degradation was determined using spectrophotometer (UV-Vis) at a wavelength of 412 nm. In the experiment it was obtained that the maximum concentration of PNP that still tolerated by Pseudomonas aeruginosa is 100 ppm tolerance, both in LB and MM. The optimum growth temperature was reached at 30oC. The presence of glucose and NH4Cl were still needed to support the growth of Pseudomonas aeruginosa, in this case as a source of carbon and nitrogen text
