Crosses between Arumanis 143 (AR 143) with other red mango varieties has been done by Research Institute of Tropical Fruit Plants. The genetic information among mangoes germplasm is considered as important and is needed to produce mangoes with higher economic value. This research was conducted to study F1 progenies phylogenetic relationship from crosses between AR 143 and some red mango varieties using ITS region nuclear ribosomal DNA. Phylogenetic tree was constructed based on sequence variation of ITS regions of 33 samples consisting of 10 parent mango plants (1 AR143 and 9 varieties of red mango), 22 F1 progenies from crosses between parent mango, and 1 outgroup samples from mango sistergroup (Mangifera indica L.). Genomic DNA were extracted and amplified using ITS primers and sequenced using direct sequencing method. Sequencing results were analyzed and aligned with ClustalX software. Furthermore, the results were analyzed using PAUP*4.0b10 to construct phylogenetic trees using maximum parsimony method. The results showed that 621 characters with 368 informative characters was obtained in the formation of the phylogenetic tree. This results also suggested that the ITS region can be used as a good genetic marker to study phylogenetic relationship in these samples. Phylogenetic analysis showed that AR 143 variety was separated from red mango varieties and F1 progenies from crosses between parent mango. This separation indicates that the evolution might occurred between AR 143 variety and red mango varieties, and have different areas of origin. Molecular phylogenetic studies based on ITS region provides information to produce new mango varieties with highest quality through crossbreeding. This promising method can be used in mango cultivation practice and also to fulfill of mango fruit consumption. text
