One banana flower-visiting insects suspected spread of disease vectors are insects of the order Hymenoptera Trigona sp. Trigona sp insect survival when bacterial infection is caused due to an active mechanism of the body's immune system. The immune system of insects consists of cellular and humoral immune system, which in general will involve activation of the immune system's role blood cells (hemocytes). Based on this, the goal of this research is to determine how the activation of the immune system of the insect vector Trigona sp. when infected with the blood of disease-causing bacteria. Research conducted at the Laboratory of Biological Insect, School of Biological Sciences and Technology, Institute of Technology Bandung, in January-August 2016. The test was obtained from Livestock Insects Trigona Kampung Cibeusi, Ciater Subang, West Java. Research conducted using a completely randomized design (CRD) with one single factor, namely the number of bacterial cells. Infections microbial pathogens (Blood disease bacterium and Escherechia coli) were conducted using natural infection (natural infection) through a system of oral infections, the number of cells of different bacteria, namely: 4.8 x 109 (P1), 9.5 x 109 (P2), 14.2 x 109 (P3), 19 x 109 (P4). Before infected, test insects were fasted for 2-3 hours with the goal of maximizing the test insects feeding process. Be infected bacterial culture was mixed with 5% sucrose solution. Old infections varied : 4, 8, 12 and 24 hours. The survey results revealed that there are five types of hemocytes on Trigona sp larvae, which plasmatosit, prohemosit, sperul cells, granulocytes and oenositoid, with the highest number of hemocytes is granulocytes prohemosit 44% and 26%. The survey results revealed that there are five types of hemocytes on Trigona sp larvae, which plasmatosit, prohemosit, spiral cells, granulocytes and oenositoid, with the highest number of homicides is granulocytes prohemosit 44% and 26%. 8 iv hours old infections, bacterial infections BDB: 0.33 units / mg protein in treatment P3, E. coli infection: 0.26 units / mg protein in the treatment of P2 and P4. 12 hours old infections, bacterial infections BDB: 0.18 units / mg protein, E. coli infection: 1.87 on P2 treatment. 24 hours old infection, infection BDB: 025 units / mg protein, E. coli infection: 1.14 on the treatment P1. Test the antimicrobial activity of the extract supernatant Trigona sp tested by the method of pitting, shows the power of 1.9 cm minimum inhibitory to bacteria BDB and showed no inhibition minimum on E. coli bacteria. The conclusion of this research that there are 5 types of blood cells (hemocytes) insect Trigona sp namely, prohemosit, plasmatosit, instead, cell spherule and journalist. BDB E. coli bacterial infection and induce humoral response Trigona sp characterized by increased activity unit phenoloksidase and their antibacterial peptide synthesis. text |
