Summary:Chilli pepper (Capsicum annuum) is one of the major commodity in Indonesia, its consumption level was reach 1.5 ton/capita/year. On the other hand, the chilli pepper production was decreasing each year caused by PYLCV infection which is transmitted by Bemisia tabaci. To overcome this problem and increase chilli pepper production, virus resistant plant was developed. The conventional method to obtain a good candidate of virus resistant plant has been done by screening using Bemisia. However, this method is unspecific on virus transmition and labour intensive. The screening method using Agrobacterium tumefaciens offers more specificity than Bemisia. In order to increase its effectiveness, two origin of replication was added to the viral genome. Based on these information, this study focus on PYLCV genome construction which has two origin of replication via several methods which are partial tandem repeat, PCR walking, complementary double-strand synthesis - PCR walking, synthetic double-strand with phosphate addition, PCR walking from one side of restriction and synthetic genes. The result showed that the construction of the PYLCV genome (DNA-A and DNA-B) consisting of 2 origin of replication was succeeded through the synthetic gene method.
