Solanesol is a secondary metabolite compound that can be extracted from tobacco plants. This compound has very important benefit values in the synthesis of coenzyme Q10 (CoQ10). Coenzymes are well-known compounds that have many benefits for the health and cosmetics industries. This study aims to determine the effect of solanesol content in increasing CoQ10 production. CoQ10 production in this study used Agrobacterium tumefaciens as its producer. Microbes are an effective and economical CoQ10 production agent. Solanesol precursors were obtained from extraction of tobacco leaves using heat-reflux method. CoQ10 production by bacteria using yeast extract broth medium with various addition of solanesol concentrations of 0 g/L, 0.5 g/L, 1 g/L, 1.5 g/L, 2 g/L, and 2.5 g/L. Yield of crude solanesol extract from tobacco leaves is 0.76-1.97%. The results of this study indicate that solanesol precursors can increase CoQ10 production as well as bacterial cell biomass. The most optimal concentration of the addition of solanesol precursor is 2.5 g/L. The dry cell weight, CoQ10 production, and specific CoQ10 content of this variation are 2.82 g, 12.02 g/L, and 4.27 mg/g-DCW. The addition of precursors can increase CoQ10 production by 217%. text |
