Summary:Blood glucose levels are regulated by insulin and glucagon hormones produced by pancreas. Hyperglycemia is a condition that higher blood glucose levels than set point. Conditions of hyperglycemia can increase oxidative stress that plays a role in the development of diabetes mellitus. Pandan wangi leaves contains antioxidants that can bind to free radicals thus might be able to reduce the development of diabetes mellitus. This study aimed to determine the effect of pandan wangi leaves ethanol extract on blood glucose levels and pancreatic histology of mice induced into hyperglycemia compared with glibenclamide. A total of 30 mice Swiss Webster males were divided into 6 groups. P0 group was the control group (solvent control) and 5 groups of P1, P2, D1, D2 and D3 is the treatment groups were induced into hyperglycemia using 150 mg / kg Streptozotozin (STZ) intraperitoneally. P1 is the control of hyperglycemia group, P2 is a comparative control using glibenclamide dose of 0,65mg / kg, D1, D2 and D3 treatment group hyperglycemia treated with ethanol extract of pandan wangi leaves with each dose of 210, 420, and 630 mg / kg BB. Measurement of blood glucose levels on days 0, 7, 14 and 21 using a glucometer. On the 24th day mice sacrificed and isolated pancreas. Pancreatic dehydrated in a series of alcohol solution continued with the process of clearing with a series of alcohol and xylol mixture. Before entering the embedding process, the sample is infiltrated in paraffin three times. Samples were planted sliced using a microtome. Pancreas stained using Hematoxylin Eosin staining (HE) and the percent damage of the Langerhans islets calculated with image-J software. The results showed that the group of mice given the extract ethanol hyperglycemia pandan wangi leaves have blood glucose levels were significantly lower and better glucose tolerance than the group P1 with significance value 0,00-0,006. Damage to the islets of Langerhans of the pancreas in the group treated with pandan wangi leaves ethanol extract showed the most damage was lower than the D3 (13.06%) and highest in the group P1 (59.01%). From the results of this study concluded that ethanol extract pandan wangi leaves have activity of anti-hyperglycemia indicated by a decrease in blood glucose levels and a reduction in the percentage of damage to the islets of Langerhans in mice induced hyperglycemia.
