Summary: | Compost production derived from vegetable waste can help reduced organic waste in environment and also increase soil fertility. This study was done to produce compost from vegetable waste using Marasmius sp. and isolated indigenous microorganism. In this study, cellulase activity, amylase activity, laccase activity, and humic substances, and also the effect of compost on Lactuca sativa growth were done. Marasmius sp. was inoculated on substrate that was consisted of corn (50%), vegetable waste (25%), rice (20%), NPK (4%), and glucose (1%), with moisture varied from 50%, 75% and 90%. During incubation, tests were done at 0-day, 3-day, 5-day, and 7-day. Amylase and cellulase activity were observed by doing Dinitrosalisilic acid (DNS) method. Laccase activity was observed using 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulhoric acid) (ABTS). Humic substances were extracted using NaOH 0.1N, and compost effect on Lactuca sativa growth was observed by varying the soil media, soil with compost, soil with NPK,and soil. The result showed that incubation time with highest humic substances content was 5 days (moisture content 75%), with humic acid content 135 mg/gram sample and fulvic acid content 83 mg/gram sample. Crude enzyme of amylase has the highest activity, 79 U/ml, at day-3 (moisture content 90%). Laccase has the highest activity, 5.5 U/ml, at day-7 (moisture content 90%), and cellulase has the highest content of 74.17 U/ml at day-7 with moisture content 90% and 75%. Leaf and root length were 3.4cm and 5.4cm (soil with compost), 2.38cm and 2.88cm (soi with NPK), and 2.1cm and 2.24cm (soil). In conclusion, the best incubation time for composting by Marasmius sp. and indigenous microorganisms was 5 days with moisture content 75%, and the best growth of Lactuca sativa was observed on L. sativa that was grown in soil with compost.
