Summary:Breast cancer is the second highest prevalence a cancer disease in Indonesia, with the number of new cases and deaths continues to increase. Kirinyuh (Austroeupatorium inulaefolium (Kunth) R.M. King & H. Robinson) is a shurb plant with a very large population. Traditionally, it is used as a treatment for wounds and cancer. Information about anticancer from kirinyuh is still very limited. This study aims to assess the potential of cytotoxic activity of kirinyuh on MCF-7 and T-47D breast cancer cells. Tests carried out were phytochemical screening, Brine Shrimp Lethality Test (BSLT) and 3- (4,5-dimethylthiazol-2-il) -2,5-dipheniltetrazolium bromide (MTT) assay. The parameters of the test results from BSLT is LC50 and MTT assay is IC50. Leaves at levels 1-3 (A) and levels 4-6 (B) from shoots and stalks kirinyuh (C) refluxed using 96% ethanol to obtain leaf extracts level 1-3 (EA), leaf extract level 4-6 (EB) and stalk extract (EC). The extract was tested with BSLT, extract which was actively fractionated, obtained water fraction (FA), ethyl acetate fraction (FEA) and n-hexane fraction (FNH). Cytotoxicity testing of extracts and fractions was carried out using MTT assay. Phytochemical screening results from EA, EB and EC contains tannin, steroids/triterpenoids, flavonoids, and phenols. The BSLT results showed that the EB has the smallest LC50 value of 12.86 ?g/mL. MTT assay results showed the smallest IC50 value of the extract and the fraction on MCF-7 cells were EA 204.96 ?g/mL and FEA 205.43 ?g/mL, and the smallest IC50 value of extract and fraction on T-47D cells were EB 217.03 ?g/mL and FEA 192.36 ?g/mL.
