Summary:Biopigments from marine microalgae Navicula sp. have potential use for an active ingredient of sunscreen due to showing ability to absorb UV light. The present research aimed to determine the value of sun protection factor (SPF) biopigments of Navicula sp. The research methodology included production of Navicula sp. biomass in seawater medium enriched with urea, phosphate, silicate, and iron; isolation of biopigments from dried biomass in acetone solvent and initial characterization of crude extract by thin layer chromatography (TLC) with n-hexane:acetone (7:3) (v/v) as eluent; purification of biopigments by 60G silica column chromatography with a gradient concentration of n-hexane:acetone 4:1, 3:2, 2:3, and 1:4 (v/v); characterization of biopigments by UV-Vis spectrophotometer; and determination of SPF value of crude extract and purified biopigments by UV-Vis spectrophotometer at 290, 295, 300, 305, 310, 315, and 320 nm. The results showed that the average of Navicula sp. cell density cultured for 7 days was 2.03±1.01 g/L. The average of water content in wet biomass of Navicula sp. was 81.52±1.11%. Six pure biopigments were purified from crude extract of Navicula sp. biopigment, and each was identified as ?-carotene, pheophytin a, chlorophyll a, diatoxanthin, diadinoxanthin, and fucoxanthin. The value of SPF crude extract (0.40 mg/mL), ?-carotene (5.62 mg/mL), pheophytin a (5.80 mg/mL), chlorophyll a (5.35 mg/mL), diatoxanthin (5.37 mg/mL ), diadinoxanthin (5.09 mg/mL), and fucoxanthin (5.22 mg/mL) was 5.29±0.36, 3.78±0.60, 4.05±0.37, 3.28±0.37, 2.54±0.16, 3.09±0.10, and 6,04±0,35. Thus, the ability of crude extract of Navicula sp. biopigment as an active ingredient of sunscreen was better than that of individual purified biopigment.
