Summary:Indonesia has a very large sea area, possessing a high diversity of microalgae. A fast and precise method is needed to determine the species of microalgae. Forensic study is used to identify species of microalgae based on cell morphology and genetic information. The purpose of this research was to characterize the morphology of microalgal cells and to determine and analyze the nucleotide sequence of LSU D2 / D3 and 18S rRNA V4 genes of two microalgae from Indonesian tropical ocean. Research methodology included activation and cultivation of pure microalgae sample, morphological characterization of microalgae cells under an optical microscope and Scanning Electron Microscope (SEM), and the determination and analysis of the nucleotide sequence of LSU D2 / D3 and 18S rRNA V4 gene. The results showed that the PLO cells are round and greenish yellow, while the NWPL cells have the shape like a "nut shell" with the colour of brownish yellow. Analysis of the nucleotide sequence of LSU D2/D3 and 18S rRNA V4 gene indicates that PLO has the greatest similarity with genus Pseudobodo, while NWPL has the greatest similarity with genus Psammodictyon.
