This study aimsed to determine total fatty acid content from lipid of the tropical marine P. cruentum. The methods included cultivating P. cruentum cells in Walne medium in seawater, harvesting microalgae biomass, extracting lipid, converting total fatty acid into fatty acid methyl ester (FAME), and characterizating FAME by the method of TLC and GC-FID. The results showed that the average cell density of P. cruentum cultured with an initial cell density of 600,000 cell/ mL for 9 d was 9,078,600 cell/mL with a biomass obtained of 25.79±0.85 g, of equivalent to the biomass productivity of 0.96±0.24 g/L/d. The lipid content of P. cruentum was 9.63% w/w dry weight. P. cruentum lipid was identified as cholesterol esters, triacylglycerols, monoacylglycerols, diacylglycerols, free fatty acids and phospholipids. Total fatty acids identified in the P. cruentum lipid was butyric acid (C4:0), capric acid (C10:0), undecane acid (C11:0), lauric acid (C12:0), tridekanoic acid (C13:0), stearic acid (C18:0), arachidic acid (C20:0), tricosanoic acid (C23:0), erucoic acid (C22:1n9), nervonoic acid (C24:1), EPA (C20:5n3), and eikosadienoic acid (C20:3n6). Thus, P. cruentum is potential to be developed as a source of essential fatty acids. text
