Research on Morus which plant samples developed by using tissue culture techniques showed this plants produce phenolic compounds mainly Diels-Alder adducts, in addition to stilbenes, 2-arylbenzofurans, and flavonoids. Some of these secondary metabolites showed important and diverse bioactivity, such as anticancer, antimicrobial, antihypertensive, and antioxidants. Phytochemistry study on shoot culture of Morus shalun have never done before so isolation and characterization of secondary metabolite from Morus shalun shoot culture were implemented in this research. Shoot culture prepared with tissue culture technique in a MS (Murashige-Skoog) liquid medium with the addition of BA (benzyladenine) hormone 2 µM. Then, isolation and purification of methanol extract from shoot culture Morus shalun done by various chromatographic techniques yielded three pure compounds. Characterization using NMR spectroscopic data demonstrated that three of these compounds consist of a flavan prenylated derivatives compound namely 7,2’-dihidroksi-4’-metoksi-8-prenilflavan, an 2-arylbenzofuran derivatives compound with cyclic prenyl group namely moracin G, and a Diels-Alder adduct derivatives compound between chalcon with dehydroprenyl-2-arylbenzofuran namely chalcomoracin. The 7,2’-dihydroxy-4’-methoxy-8-prenylflavan is new flavan derivates compound on Morus culture, but it has been reported from Morus natural plant. Meanwhile, two other compounds that is moracin G and chalchomoracin has been isolated from Morus culture. In cytotoxicity toward murine leukemia P-388 cell, the isolated compounds as 7,2’-dihydroxy-4’-methoxy-8-prenylflavan and moracin G have cytotoxicity not active with IC50 5,8 and 5,5 µg/mL, but chalcomoracin showed very high activity with IC50 1,6 µg/mL. text |
