Traditionally, Tempuyung, Sonchus arvensis L., has been used to lower the blood pressure, remove kidney stones and overcome boils and in preclinical study, it has shown having antimicrobial and cardiovascular protective activity. Binahong, Anredera cordifolia (Ten) V. Steenis, has been used traditionally to heal wound and treat diabetes mellitus, tuberculosis, rheumatic, gout and hypertension and in preclinical study, it has been shown to have antihyperlipidemia activity. Recently, research of the combination of ethanol extracts of tempuyung’s leaves and binahong’s leaves has shown the efficacy as antihypertensive and antihyperuricemia agents. The toxicity data of each ethanol extract of both plants represented on LD50 is more than 5 g/kg bw and safe for subchronic use, but there is no sufficient data for subchronic safety profile of the combination. The aim of the study is to obtain a subchronic usage safety profile of the combination. Tests were carried out based on PerKa BPOM No. 7 year 2014 which tested on three groups of doses which doses were the combination of 50-50 mg/kg bw, 200-200 mg/kg bw and 500-500 mg/kg bw. Based on the behavioral parameters, body weight, relative organ weight, urine, hematology, clinical biochemistry, macropathology and histopathology analysis, the combination of both extracts showed no significant difference compared to the control group and there was no significant difference after the termination use of the extract combination at high doses. In conclusion, the use of the combination of ethanol extract of tempuyung leaves and binahong leaves up to the dose of 500-500 mg/kg bw was safe.
