Castor seed oil contains fatty acid triglycerides which act as emollient to maintain skin moisture and vitamin E as antioxidant to fight free radicals. The purpose of this study was to produce stable O/W topical microemulsion of castor seed oil. The optimization formula of castor seed oil microemulsion was carried out using castor seed oil as the oil phase, surfactant (Cremophor EL, Cremophor RH 40, and Tween 80), cosurfactant (glycerin and PEG 400), methyl and propyl paraben, propylene glycol, and distilled water with various concentration. Evaluation of microemulsion included organoleptic assessment, specific gravity, viscosity, stability test in room temperature, accelerated stability test, freeze-thaw test, centrifugation test, skin irritation test, ocular irritation test, antioxidant activity test, and antiwrinkle activity test using Video- Dermatoscopy. The parameter of globule size, polydispersity index, and pH was used to obtain the physical stability of the microemulsion. The best microemulsion formula of this study was sucessfully constructed by using 24% of Cremophor EL, 16% of PEG 400, 5% of castor seed oil, 0.18% of methyl paraben, 0.02% of propyl paraben, 10% of propylene glycol, and 44.8% w/w of distilled water. Microemulsion obtained was clear yellowish in colour and had distinctive smell, with globule size 258.37 ± 21.41 nm, polydispersity index 0.294 ± 0.06, pH 5.66 ± 0.01, density 1.05, viscosity at 50 rpm was 100.67 ± 4.19 cPs, and potential zeta 0.06 mV. The results of stability test showed that microemulsion was stable for 28 days of storage in room temperature and 40?C. Microemulsion was also stable during six cycles of freeze-thaw test. The centrifugation test with rotation speed of 3800 rpm for 5 hours showed that the preparation was stable without any separation phase. Microemulsion was not irritant, both in skin and eye irritation tests. The antioxidant activity test was carried out using the DPPH method . The antioxidant activity of castor seed oil and microemulsion shown by IC50 values were found to be 71,41 ?g/mL and 50,82 ?g/mL respectively. Antiwrinkle activity test of castor seed oil microemulsion for 28 days of examination showed that there was no significant reduction in wrinkles statistically (p?0,05). text |
