Summary:Lemongrass oil is an essential oil obtained by steam distillation of Cymbopogon flexuosus Steud. leaves. Lemongrass oil has antioxidant activity that can scavenge free radicals on the skin so that it can be used as an anti-wrinkle to prevent premature aging. This study aimed to make and evaluate a microemulsion formula contain lemongrass oil. Optimization of lemongrass oil microemulsion formula was prepared by using surfactants (Cremophor RH 40, Tween 80), cosurfactants (PEG 400, glycerin), preservatives (methyl paraben and propyl paraben), propylene glycol as paraben solvent, and water. Evaluation of microemulsion was carried out on the physical stability of the product, antioxidant test for lemongrass oil and lemongrass oil microemulsion, acute dermal irritation test, eye irritation test, and antiwrinkle test with video-dermatoscopy instrument. Microemulation of lemongrass oil was successfully constructed by using 15% Cremophor RH 40, 10% PEG 400, 7% lemongrass oil, 10% propylene glycol, 0,18% methyl paraben and 0,02% propyl paraben. The microemulsion had clear and light yellow appearance with a distinctive smell of lemongrass; globular size was 223,47 ± 30,72 nm; polydispersity index was 0,31 ± 0,05; pH was 5,44; potential zeta was 0,00 mV; specific gravity was 1,037; and viscosity was 73,33 cP. There was no phase separation or precipitation occurred during the centrifugation test. Antioxidant test using DPPH method produced IC50 of lemongrass oil was 6.084 ± 0,059 µg/mL and IC50 of lemongrass oil microemulsion was 3,119 ± 0,124 µg/mL. The result of statistical analysis with one-way ANOVA in the room temperature stability test, accelerated stability test, and freeze-thaw stability test showed that the globular size of microemulsion did not change significantly (p ?0,05) after 4 weeks of storage. The dermal acute irritation test and eye irritation test showed that the microemulsion did not irritate the skin and eyes. The result of statistical analysis with one-way ANOVA in antiwrinkle test showed that wrinkle did not change significantly (p ?0,05) after 4 weeks of usage.
