Therefore, development for another effective, safe, and affordable alternative therapy from herbal medicine is necessary. Some of potential plants effective for antibacteral therapy are sappan wood (Caesalpinia sappan L.) and avocado seed (Persea americana Mill.). The aim of this study was to determine antibacterial activity, Minimum Inhibitory Concentration (MIC), Minimum Bactericidal Concentration (MBC) of sappan wood and avocado seed ethanolic extracts, and antibacterial combination activity of extracts against Escherichia coli and Staphylococcus aureus. This study began with antibacterial activity screening test of extracts using agar diffusion with paper disk, and continued with MIC and MBC determination using broth microdilution method. Determination of antibacterial combination activity of extracts was done using Checkerboard and Time-Kill Curve methods. Results showed that sappan wood and avocado seed ethanolic extracts had stronger antibacterial activity against S. aureus than E. coli. MIC of sappan wood and avocado seed ethanolic extracts against E. coli were 1024 dan 8192 µg/mL, respectively, and against S. aureus were 256 µg/mL. MBC of sappan wood and avocado seed ethanolic extracts against E. coli were >8192 µg/mL, and against S. aureus were 256 µg/mL. Additive combination were obtained from test results against both test bacterias, with FICC index of extract combination against E. coli and S. aureus were 0.75 and 1, respectively. text
