Summary:Uric acid is the result of purine metabolism produced by the body. When the level of uric acid in the blood increased, this condition can be called hyperuricemia. If this uric acid settled in the joint there will be inflammation called gout. One of the recommendations for treatment of gout is xanthine oxidase inhibitors. Papaya (Carica papaya L.) is one of the plants that was known to be able to inhibit the xanthine oxidase enzyme in vitro. The compound that responsible in inhibiting xanthine oxidase were not known. This study aimed to determine the xanthine oxidase enzyme inhibitors activities of n-hexane, ethyl acetate, and ethanolic extract of papaya leaves, and to isolate, characterize, identify of inhibitor xantin oxidase active compound. Crude drug from papaya leaves was extracted by maceration method with different polarity of solvents which were n-hexane, ethyl acetate, and ethanol. Furthermore, the extracts were tested for their inhibitory activity against the xanthine oxidase enzyme. The activity of xanthine oxidase enzyme inhibitor was carried out using UV-Vis Spectrophotometry instrument. Fractination was carried out by vacuum liquid chromatography using several combinations of solvents with increasing polarity, which were n-hexane, ethyl acetate, and methanol. The selected fraction was purified by preparative thin layer chromatography, so that isolate A was obtained. The purity test of isolate A was carried out using thin layer chromatography method with three different mobile phases with increasing polarity so that pure isolate A was obtained. Isolate A was then tested for inhibitory activity against the xanthine oxidase enzyme. Isolate A was characterized by specific reagent and spectrophotodensitometry thin layer chromatography. The ethanolic extract of papaya leaves showed a stronger activity of xanthine oxidase inhibition than the other extracts. Percent inhibitor of ethanolic extract with a concentration of 1000 µg/mL was 41.54 ± 0.55% against xanthine oxidase. Isolate A showed the maximum wavelength at 263 nm, it was predicted as flavonoid with phenol substituen in the structure. Percentage of inhibition of isolates A at 230 µg/mL was 62.67 ± 0.27%.
