Summary: | Thyme has been known empirically and scientifically to have antimicrobial activities, especially
against Streptococcus mutans, which is responsible for tooth decay in humans. However, its use
as an active ingredient in formulation for dental health has not been found. The purpose of this
research was to develop a toothpaste formulation containing thyme extract as an antimicroba
and also as a solvent. Thyme extract was prepared by heating thyme in boiling aquadest, then
steam was passed through a cooler, and the extract was collected in a closed container. The
extraction process was carried out until the ratio extract to thyme is 3:1. Thymol contens in thyme
extract was determined using TLC densitometer. The thymol content in thyme extract was 83,49
ppm. Thyme extract was tested for its antibacterial activity by determining the diameter size of
growth inhibitory of Streptococcus mutans using disk diffusion method and the minimum
inhibitory concentration (MIC) using microdilution method. Based on the results, thyme extract
gave an activity of growth inhibition of Streptococcus mutans with a diameter 16,8±5,6 mm and
MIC 256 ppm. The toothpaste formula consists of 30,1% kaolin, 15% glyserin, 7,5% propylen
glykol, 2% titanium dioxide, 3% Na CMC, 2% sodium lauryl sulfat, 1% menthol, 0,2% sodium
saccharin, 0,15% methyl paraben, 0,05% propyl paraben, and extract up to 100%. Toothpaste was
tested for in vitro antimicrobial activity in the form of growth inhibitory of Streptococcus mutans
colonies, which were quantified using a colony counter. Based on the results, toothpaste contains
thyme extract gave 36,71% growth inhibiton of Streptococcus mutans.
