Summary:Katuk (Sauropus androgynus (L.) Merr.) is a plant that is widely used as traditional medicine. In Indonesia, katuk leaves are used as an ASI booster (laktagoga), treatment of boils, ulcers, zits and, fever. Based on research, one of secondary metabolites groups that found in katuk leaves is flavonoids. Flavonoids have been widely used in the medical field because it have various activities such as anti-inflammatory, antioxidant and antibacterial effects, so this study aims to isolate, characterize, and identify flavonoid compounds that contained in katuk leaves. The crude drug of katuk leaves were extracted by maceration method using methanol solvent. Methanol extract was monitored using the thin layer chromatography (TLC) method with the citroboric spray reagent. Chlorophyll separation is done by adding hot water to the methanol extract and filtering it out. The filtrate obtained was fractionated by liquid-liquid extraction method using a solvent with multilevel polarity namely n-hexane and ethyl acetate, so that three fractions were produced, namely n-hexane fraction, ethyl acetate fraction, and water fraction. The three fractions were monitored by TLC method. In the ethyl acetate fraction there were at least four flavonoid spots and in the water fraction there were at least one flavonoid spot, whereas the n-hexane fraction was not observed of flavonoid spot. Furthermore, the ethyl acetate fraction was sub-fractionated using preparative TLC method. Subfraction which is containing flavonoid was purified by preparative TLC method, then purity was tested by two-dimensional TLC and single development TLC with three different mobile phases. Isolates were characterized by spectrophotodensitometry-TLC, ammonia vapor spray reagent, and two-dimensional paper chromatography. Isolate is thought to be a compound of flavonol aglycone with an OH group free at C3.
